This Thursday (October, 29th) at 3pm, Ines Simoes, a PhD student from the Laboratory of Mitochondrial Biology and Metabolism, will present her work entitled: The role of hypercaloric diets in the development and progression of Non-alcoholic fatty liver disease

Abstract

Non-alcoholic fatty liver disease (NAFLD) is characterized by the development of steatosis, which can progress to inflammatory Non-alcoholic steatohepatitis (NASH) and may culminate with chronic liver failure. The progression of steatosis towards NASH has been described to be associated with mitochondrial‐induced reactive oxygen species production and to mitochondrial dysfunction. However, it is still not clarified the sequence of events that link mitochondrial-redox alterations to the pathophysiology of NAFLD. Therefore, in my PhD research project, I have hypothesized that the onset of a pro-oxidant state act as the trigger to mitochondrial and hepatic-related alterations that may drive by a non-reversible way the disease progression into NASH. Using an in vivo NAFLD mice model, I have addressed the distinct role of hypercaloric diets (fat, sucrose or combined fat and sucrose) in an early stage of NAFLD. In fact, new insights about the mechanisms governing hepatic oxidative damage were provided. Furthermore, I have investigated the specific end-points for mitochondrial dysfunction that govern NAFLD progression along time.

We will most probably meet via a new platform, I will send the instructions at the beginning of this week.

Project title:

Project is carried out within the MAB/IRA programme of the Foundation for Polish Science

Scope of the project:

Synaptic plasticity of appetitive learning

The research will focus on synaptic adaptations that occur during the formation of appetitive memory – the memory about pleasurable events. In this project, using electrophysiological recordings and in vivo optogenetics in mice, we plan to define the role of silent synapses during the acquisition and retrieval of appetitive memories. Silent synapses are immature contacts, that represent the brain’s increased capacity to learn. In the amygdala – a key brain structure responsible for appetitive and aversive learning we will study the formation and fate of silent synapses in response to natural and addictive rewards.

Postdoctoral candidates are welcome to propose their own research project, that would be complementary to Dr. Beroun’s research.

Key responsibilities of the Postdoctoral Researcher include:     

  • Planning and conducting experimental work
  • Data analysis
  • Manuscript preparation
  • Supervision of PhD students

Job starting date: December 2020

Scope of the project:

To study aberrant synaptic plasticity that underlies diseased mind

Knapska lab research is focused on investigation of neural representations of emotional states in interacting animals and humans. Using behavioral models of socially transferred positive and negative emotions as well as high-throughput behavioral measures of social interactions of their own design (Eco-HAB system, Puscian et al., eLife, 2016), neuronal tracing and optogenetics, as well as brain imaging, they characterize neuronal circuits and cellular mechanisms underlying social interactions of humans, rats and mice and validate tissue-specific therapeutic interventions using mouse models of synaptopathies relevant to autism spectrum disorders (Meyza et al., Neurosci Biobehav Rev., 2017). Postdoctoral candidates are expected to propose their own research project, complementary to these studies.
